As a player, Slot began his career at his hometown side VV Bergentheim. Afterward, he moved to FC Zwolle and played there for seven years as a goal-scoring attacking midfielder.
In 2002, he joined NAC Breda and finished fourth in the Eredivisie in his first season. He moved to Sparta Rotterdam in the summer of 2007 and played until 2010. During his time at Rotterdam, he spent a year of a loan spell at FC Zwolle before moving on a permanent deal in 2010.
Arne ended his football career at Zwolle and announced his retirement in 2013. He has appeared in over 400 games in his senior career and has scored 100 goals.
